Доказите сугерираат дека намаленото ниво на SMN (survival motor neuron) протеин може директно да влијае врз клетките и ткивата во периферијата.2,5,6

Поради делеција и/или мутација на SMN1 генот постои недоволно ниво на SMN протеин низ целото тело кое предизвикува СМА.2

Спиналната мускулна атрофија е асоцирана со широко распространети компликации и во централниот нервен систем и на периферијата.7,8
